The fixation of human tissues with formalin, and their subsequent embedding in paraffin, has been a routine method of preserving surgical samples for many years. But these extensive resources are rarely used in microarray-based gene expression or chromosome alteration studies, because nucleic acids isolated from FFPE sources are typically degraded.

Expression Analysis has partnered with NuGEN Technologies to optimize their WT-Ovation™ FFPE System. This
whole transcriptome amplification solution uses NuGEN’s proprietary Ribo-SPIA™ technology to generate biotin-labeled hybridization targets from small and degraded RNA samples that perform well in microarray expression profiling studies.

EXPRESSION PROFILING USING RNA ISOLATED FROM FFPE TISSUES
RNA samples from FFPE specimens are amplified and labeled using the WT-Ovation
™ FFPE System. Depending on the tissue procurement, fixation method and length of FFPE storage, a high percentage of transcripts, up to 50%, can be detected.

RNA AND DNA ISOLATION FROM FFPE TISSUES
Expression Analysis uses the FormaPure Isolation Kit from Agencourt Biosciences to simultaneously isolate RNA and DNA from 10-micron sections of FFPE tissue. Although the nucleic acids are often degraded, they can be used in microarray and QRT-PCR assays.
