The integrity of the nucleic acid starting material can profoundly affect the quality of microarray data. Expression Analysis offers nucleic acid isolation services which can be requested alone or in conjunction with other services. RNA ISOLATION
Expression Analysis offers RNA isolation services from cell pellets and tissues. Solid tissues are usually disrupted and homogenized in a bead-based instrument. Total RNA is isolated using QIAGEN products, including on-column DNase treatment, with some tissue-specific modifications. One of the most critical parameters influencing RNA integrity is the immediate stabilization of specimens during the procurement procedure. For tissue and cell pellet sources the two most effective methods are flash freezing and immediate transfer to a commercially available stabilization matrix, such as RNALater from Ambion.
To date we have performed RNA purifications from a wide variety of tissue and cell sources including adipose tissue, bone marrow aspirates, brain, cervix, flow cytometry separated cells, heart, jejunum, keratinocytes, liver, lung, mammary, pancreas, PBMC pellets, and skin.
DNA ISOLATION
Expression Analysis offers genomic DNA isolation services from cell pellets and tissues, including blood specimens collected in PAXgene DNA tubes. Solid tissues are usually disrupted and homogenized in a bead-based instrument. Expression Analysis uses the FormaPure Isolation Kit from Agencourt Biosciences to simultaneously isolate RNA and DNA from 10-micron sections of FFPE tissue.
